چکیده

Methods An isolated model of the rat small bowel (1) was used. Intestines were stimulated with a 0.5 nmol PAF bolus via the mesenteric artery alone (PAF, n=5) or after pretreatment with IBMX (100μM) and forskolin (0.5μM) for 20 min (PAF+PDE/AC, n=4) to increase intracellular cAMP by inhibition of phosphodiesterase (PDE) and stimulation of adenylate cyclase (AC). The pressure responses, the vascular fluid loss and the transfer of FITC-labeled vascular dextran were monitored. cAMP was measured in PAF stimulated and untreated intestines (n=5) 3 min after PAF-stimulation or at the equivalent time point.

Role of platelet-activating factor in Chinese hamster ovary cell responses to cholera toxin.

Cholera toxin (CT)-induced intestinal secretion and Chinese hamster ovary cell (CHO) elongation involves cyclic adenosine monophosphate and protein synthesis-dependent prostaglandin formation. We previously reported inhibition of CT-induced intestinal secretion and CHO elongation by platelet-activating factor (PAF) receptor antagonists and secretion of PAF by human intestinal epithelial cells e...

Platelet-activating factor increases mucosal permeability in rat intestine via tyrosine phosphorylation of E-cadherin.

1. Platelet-activating factor (PAF), an inflammatory mediator, plays an important role in mediating intestinal injury. However, it remains unclear whether PAF has a function in the intestine. The production of PAF by normal intestine and by unstimulated intestinal epithelial cell lines suggests that PAF may have a regulatory function in the normal bowel. 2. In this study we investigated the rol...

Protective Role of Rheum tanguticum Polysaccharides 1 on Radiation-Induced Intestinal Mucosal Injury

The protective effects of Rheum tanguticum polysaccharide 1 (RTP1), which is extracted from the Chinese traditional medicine Rheum tanguticum, on radiation-induced intestinal mucosal injury was investigated. Rat intestinal crypt epithelial cells (IEC-6 cells) and Sprague-Dawley rats were each divided into control, irradiated and RTP1-pretreated irradiated groups. After irradiation, cell surviva...

The role of xanthine oxidase in platelet activating factor induced intestinal injury in the rat.

BACKGROUND Xanthine oxidase (XO) is an important source of reactive oxygen species in the small intestine. AIMS To examine the interaction of platelet activating factor (PAF), XO, and neutrophils in mediating intestinal injury in rats. METHODS Two doses of PAF were used to induce either reversible hypotension, or irreversible shock with intestinal necrosis. The activities of XO, and its pre...
